Today plasma therapy in the medical field is a new method for treatment of some diseases. Cancer treatment is part of it. In this dissertation, we investigated selective toxicity of cold atmospheric pressure plasma with using argon as feed gas, on normal & cancerous mitochondria obtained animal rat model of retinoblastoma. For this reason we determined the possible toxic alterations on mitochondrial upstream events of apoptosis such as respiratory complex2 activity & level of mitochondrial ROS, potential membrane collapse, mitochondrial swelling, mitochondrial glutathione levels, lipid peroxidation & finally release of cytochrome C were analysed. In addition all plasma physical behavior from the moment of electrical discharge was studied; including plasma spectroscopy, plasma dosing, chemical reactions, produced particles & etc .
Prior to this research, the effect of cold plasma on “in vitro” conditions was investigated just on other several tumor cell lines. But in this dissertation, experiments performed on mitochondria isolated from ocular cells. This is because of unic character of mitochondria that is the main intercellular organelle to produce energy & cellular respiration, so mitochondria could be a valuable tool to evaluate of selective toxicity of any new chemical entity or physical process (plasma radiation) leading to apoptosis in tumor cells. Based on all results cold plasma showed positive & promising results on mitochondrial upstream parameters, leading to apoptosis only on cancerous ocular mitochondria without any significant effect on normal mitochondrial control.
